چکیده

A sexual pheromone, named the protoplast releasing substance (PRS), was formed by mating type minus cells. PRS activates mating type plus cells and results in the release and fusion of protoplasts within distended conjugation-papilla in paired cells and in the release and disruption of protoplasts in unpaired mating type plus cells. In an agar barrier system, the activation of mating type plus cells was markedly inhibited by treatment with pronase (5-10 micrograms per milliliter), proteinase (100 micrograms per milliliter), and alpha-mannosidase (10 micrograms per milliliter). Trypsin (10-100 micrograms per milliliter) had no effect on the activation in the agar barrier system. The results suggest that PRS is a glycoprotein with pronase-sensitive and trypsin-insensitive structure.

A sex pheromone, protoplast release-inducing protein (PR-IP) inducer, induces sexual cell division and production of PR-IP in Closterium.

The sex pheromone protoplast release-inducing protein (PR-IP) inducer and a sexual cell division-inducing pheromone-minus (SCD-IP-minus) that mediates the sexual reproduction of the heterothallic Closterium peracerosum-strigosum-littorale (C. psl) complex were investigated in this study. Recombinant PR-IP inducer produced by yeast cells was prepared and assayed for production of PR-IP and induc...

Molecular cloning of a novel sex pheromone responsible for the release of a different sex pheromone in Closterium peracerosum-strigosum-littorale complex.

A sex pheromone, protoplast-release-inducing protein (PR-IP) inducer, of the Closterium peracerosum-strigosum-littorale complex is known to induce the release of PR-IP, from mating-type plus (mt+) cells during sexual reproduction. The purified PR-IP inducer was treated with trypsin to obtain internal peptides for determination of partial amino acid sequences. Using these sequences, oligonucleot...

Characterization and molecular cloning of conjugation-regulating sex pheromones in homothallic Closterium.

Conjugation-regulating pheromones were analyzed in homothallic Closterium for the first time. Members of the Closterium peracerosum-strigosum-littorale complex are unicellular charophycean algae in which there are two modes of zygospore formation: heterothallism and homothallism. A homothallic strain of Closterium (designation, kodama20) forms selfing zygospores via the conjugation of two siste...

Expressed sequence tags from the Closterium peracerosum-strigosum-littorale complex, a unicellular charophycean alga, in the sexual reproduction process.

We obtained genetic information on sexual reproduction of the Closterium peracerosum-strigosum-littorale complex, a unicellular charophycean alga. Normalized cDNA libraries were constructed from cells in the sexual reproduction process, and a total of 1190 5'-end expressed sequence tags were established. Since 604 of these ESTs were classified into 174 non-redundant sequences, these 1190 ESTs i...

Gene expression profiling using cDNA microarray analysis of the sexual reproduction stage of the unicellular charophycean alga Closterium peracerosum-strigosum-littorale complex.

The desmid Closterium peracerosum-strigosum-littorale complex, which is the closest unicellular sister to land plants, is the best characterized of the charophycean green algae with respect to the process of sexual reproduction. To elucidate the molecular mechanism of intercellular communication during sexual reproduction, we created a normalized cDNA library from mixed cells of the sexual and ...
